Well, if based on the age most kids finally become adults, 18, then he’s been at the guardian business for at least 180 years old. Of course, you have to add a number of years when he was a “child”, I suppose. Also, I bet there’s got to be some sort of training to become a guardian.
So, all in all, I’m lost.

I would guess around 1,850.
That is assuming he had 100 assignments before Peter, and guardians stay with their charge until age 18, adding twenty years for childhood and training

Peter seems at this point still rather new to his acquaintence with Seth, and it follows that a Guardian is assigned when a child needs it, which would rarely be at birth. Figure an average of eight years old for a child to get a guardian, and if it remains until the child becomes an adult, then Seth has been a Guardian for about a thousand years.

Let’s try limiting it down, though. (Very minor spoilers, folks.)
We know Peter’s twelve. Other Guardian children seem to have started at about four, potentially from birth, and the agreed leaving point is probably about 18.
At least a hundred means 101 or higher. We also know not every child gets a Guardian from birth. So saying exactly 100 x 18 is overstating it, most likely.
I’m assuming most children get Guardians about 6, given their imagination getting especially active then. And assuming Seth would know ~150 or ~200 children, safe to assume lower than those values.
